Do you have enabled USB debugging for your Android device ?
Go to Settings->Develop Option on it and enable USB Debugging.

See the fix SP tool flash problem thread and try EVERY option there. Also see the ultimate driver pack. Remove all drivers with usbdeview and install pdanet drivers. Done.
